Is Energesis competing with GLP-1 drugs like semaglutide and tirzepatide?

In short: No.

While Energesis is developing drugs for obesity alongside popular GLP-1 medications like semaglutide and tirzepatide, that’s where the similarity ends. We do not view GLP-1s as competitors — instead, we view them as complementary partners.

 

There are two main reasons why our strategy sets us apart from the GLP-1 class:

 

1. The Future of Obesity Treatment is Combination Therapy

GLP-1s have been a massive leap forward, helping millions lose weight. However, as their use has grown, so has our understanding of their limitations, which include:

     - Loss of lean muscle mass alongside fat loss


     - Gastrointestinal side effects like nausea and vomiting


     - Weight regain once the medication is stopped


This being said, we don’t believe these drugs need to be replaced. Instead, the future of obesity management lies in combination therapy — adding new, targeted treatments to GLP-1s to address these exact shortcomings.

Combining drugs with different biological targets allows us to maximize benefits while keeping individual drug doses (and their side effects) lower. This approach is already the gold standard in more mature areas of treatment like heart disease, hypertension, and cancer. However, effective combination therapy requires using drugs that work through entirely different biological pathways.

 

2. A Completely Different Mechanism of Action

Body weight is governed by a simple balance: calories in versus calories out.

 

     - GLP-1s target "Calories In": Current GLP-1 drugs (as well as other emerging targets like GIP, glucagon, and amylin) work primarily by increasing an individual’s feeling of fullness and suppressing appetite. Stacking one appetite suppressant on top of another likely offers diminishing returns and may compound side effects.


     - Energesis targets "Calories Out": Our approach focuses on the other side of the body weight equation — increasing energy expenditure (burning calories).

 

Because our mechanism increases calorie burn rather than suppressing appetite, our drug candidates can theoretically be paired with any GLP-1 or appetite-reducing therapy.

In our preclinical animal studies, combining our lead candidate with a low dose of a GLP-1 significantly enhanced total weight loss compared to the GLP-1 alone — while preserving muscle mass.

 

The Bottom Line

GLP-1s are a historic breakthrough, and they are here to stay. Rather than trying to displace them, Energesis aims to make GLP-1 therapies better, more tolerable, and more sustainable for patients over the long term.
